Yes. Amazon Brand Registry depends heavily on a U.S. trademark — you generally need a USPTO trademark (or a pending application, depending on Amazon's policy at the time) to enroll. So registering the trademark is the first step toward Brand Registry.

I registered a U.S. company — does that mean I have a trademark?

No. Company registration and trademark registration are two different things: your company name is your operating-entity identity, while a trademark protects the brand name/logo you sell under. They're applied for separately and can even be different names.
